Abstract:Objective This study aimed to fabricate a composite hydrogel designated as SA/CUR-GO,composed of sodium alginate(SA),curcumin(CUR),and graphene oxide(GO),followed by comprehensive characterization of its morphological architecture, physicochemical properties, cytocompatibility, and osteogenic differentiation effects. Methods SA/CUR-GO composite hydrogels were prepared using a freeze-ionotropic gelation method. The experimental groups comprised SA,SA/GO,and SA/CUR-GO.Microstructural morphology was characterized using scanning electron microscopy(SEM).Chemical structural analysis was performed employing Fourier Transform Infrared Spectroscopy(FT-IR).Mechanical properties were assessed via compression testing using a universal testing machine. For in vitro biological evaluation, mouse bone marrow mesenchymal stem cells(mBMSCs)were cultured on the surfaces of the three hydrogel groups. The detection indicators included the effects on cell viability, proliferation ability, and osteogenic differentiation potential. Results Micro-morphological analysis revealed that all hydrogels exhibited an interconnected porous network.Notably, the SA/CUR-GO hydrogel displayed increased surface roughness and significantly enhanced mechanical strength. In vitro cellular experiments demonstrated that the SA/CUR-GO composite hydrogel possessed favorable cytocompatibility and significantly promoted the osteogenic differentiation of mBMSCs. Conclusion The SA/CUR-GO composite hydrogel demonstrates outstanding cytocompatibility and remarkable ability to promote the osteogenic differentiation of mBMSCs.These characteristics make it a highly promising biomaterial, especially with broad application prospects in the fields of periodontal tissue regeneration and orthodontic treatment.
