Effect of Matrine on Proliferation Invasion and TGF-β1/Smad4 Protein Expression to colon cancer cell line SW620

Abstract:Objective To investigate the effect of matrine on proliferation,invasion and TGF-β1,Smad4 protein expression in colon cancer cell line SW620. Methods The effect of matrine on proliferative activity in SW620 cells were evaluated by CCK8 assay and colony formation assay.The effect of matrine on migration and invasion ability in SW620 cells were evaluated by Transwell assay.Quantitative analysis the effect of matrine on Ki-67,E-cadherin,TGFβ1 and Smad4 proteins expression in SW620 cell blocks by Immunohistochemistry. Results The proliferation and invasion of SW620 cells were significantly weakened after matrine treatment.TGF-β1 and Smad4 proteins expression in SW620 cells were absent,matrine had no significant effect on the proteins expression of TGF-β1 and Smad4. Conclusion Matrine can inhibit the proliferation and metastasis of SW620 cells,but the TGF-β1/Smad4 signaling pathway mechanism may be independent.
