Study on the Relationship between miRNA-146a Expression in Peripheral Blood Mononuclear Cells and Oxidative Stress Levels and Inflammatory Response in Patients with Chronic Obstructive Pulmonary Disease

Abstract:Objective The primary objective of this study was to explore the intricate relationship between the expression of miRNA-146a in peripheral blood mononuclear cells(PBMCs) of patients afflicted with chronic obstructive pulmonary disease(COPD)and both the oxidative stress levels and the inflammatory response. Methods Peripheral blood samples were procured from 52 individuals diagnosed with COPD and 52 healthy counterparts.The expression of miRNA-146a in PBMCs was detected using real-time fluorescence quantitative PCR technology.Biochemical assay kits were employed to ascertain the levels of oxidative stress markers,and ELISA was utilized to measure the levels of inflammatory response markers.Subsequently,a comprehensive comparison of the correlations among these indicators was conducted. Results In contrast to the healthy control group,a significant reduction in the expression level of miRNA-146a in the PBMCs of COPD patients was observed(P<0.05).Notably,as the GOLD grading advanced,a downward trend in the expression level of miRNA-146a became evident.The levels of malondialdehyde(MDA)in the plasma of COPD patients were markedly higher than those in the healthy control group(P<0.05),while the levels of superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)were substantially lower than those in the healthy control group(P<0.05).Additionally,the levels of interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),and interleukin-1β(IL-1β)in the plasma of COPD patients were significantly elevated compared to those in the healthy control group(P<0.05).Intriguingly,the expression level of miRNA-146a in the PBMCs of COPD patients exhibited a negative correlation with MDA levels(both P<0.05),and a positive correlation with SOD and GSH-Px levels(P<0.05).Concurrently,the expression level of miRNA-146a was inversely correlated with the levels of IL-6,TNF-α,and IL-1β(all P<0.05). Conclusion The expression level of miRNA-146a is intimately intertwined with the severity of COPD and demonstrates a significant correlation with oxidative stress levels and inflammatory response indicators.It is postulated that miRNA-146a might play a crucial role in the pathogenesis of COPD.
