1 Construction and Characterization of SARS-CoV-2 N-Ferritin Prokaryotic Expression Plasmids

Objective To construct the prokaryotic expression vector pET32a-N-Ferritin-Flag-6His by a novel ferritin vaccine platform,optimize the induction expression condition of the recombinant protein N-Ferritin,and characterize its morphological structure and stability in depth. Methods The full-length sequence of the SARS-CoV-2 N(MT108784.1)protein was connected to the N-terminus of the ferritin protein by a flexible linker(G4S)3,and the C-terminus was connected with the Flag and 6×His tags,which was inserted into the pET32a vector,and the recombinant plasmid pET32a-N-Ferritin-Flag-6His of the fusion expression protein N-Ferritin was constructed.The pET32a-N-Ferritin-Flag-6His plasmid was transformed into the BL21(DE3)competent cells,and the optimal induction condition of the recombinant protein N-Ferritin was determined by optimizing the final concentration of IPTG in the induction process,time and temperature.The induced recombinant protein N-Ferritin was purified by nickel affinity chromatography column,and the recombinant protein was verified and identified by SDS-PAGE,Western blot,transmission electron microscopy,and DLS.The stability of the recombinant protein was analyzed by SDS-PAGE and Coomassie brilliant blue staining under different freezing and thawing times and different storage temperatures. Results The recombinant N-Ferritin protein was successfully prepared,and there was a target band at 85 kDa.Transmission electron microscopy observed that the N-Ferritin recombinant protein showed a uniform spherical particle.The DLS results showed that the particle size of the recombinant protein was 18.23 nm.Repeated freezing and thawing experiments showed that the N-Ferritin recombinant protein still had good stability after repeated freezing and thawing 3-5 times and stored for 4 weeks under different temperatures(80℃,20℃,4℃). Conclusion The N-Ferritin prokaryotic expression vector was successfully constructed,and a large amount of high-purity target protein could be obtained after optimizing the induction conditions,and it has good stability.

6 Overexpression of Chemokine Receptor 2 Combined with Rat Adipose Derived Stem Cells(ADSCs)Promotes Wound Healing of Full-thickness Skin Defects

Objective To investigate the effect of rat adipose-derived stem cells(ADSCs)overexpressing chemokine receptor CXCR2 on the healing of skin defects. Methods Three SD rats were executed,the adipose tissue around the groin and epididymis was rapidly isolated,and ADSCs were extracted by enzymatic digestion and cultured to the third generation.The morphological characteristics of the cells were examined by inverted microscopy,while flow cytometric analysis was used to assess the phenotypic markers of the cells and their differentiation ability.Forty mice were selected to construct an animal model of dorsal total skin defects and randomly assigned to four experimental groups:control group(given saline treatment),adipose-derived stem cells(ADSCs)treatment group,CXCR2 intervention group,and combined treatment group(ADSCs and CXCR2 co-application).Saline,ADSCs suspension(100 μL PBS),or CXCR2(10μg/L)were injected locally at the edge and base of the trauma,respectively.The healing progress was monitored by calculating the percentage of wound area to the initial area and the tissue changes of the wounds were observed by HE staining;the serum levels of IL-1β,IL-18 and TNF-α were detected by ELISA.The serum levels of IL-1β,IL-18 and TNF-α were detected by ELISA.The data between groups were compared by ANOVA and t-test. Results Under the microscope,primary ADSCs initially appeared spindle-shaped,transitioning to circular after 2 hours,and predominantly spindle-shaped by 48 hours.The third generation of cells exhibited a vortex arrangement.The combination therapy group showed significantly accelerated wound healing compared to the model group,with healing areas notably reduced at days 3,7,14and 21(P<0.01).HE staining indicated complete wound healing and minimal inflammatory cell infiltration in the combination group,while the ADSCs and CXCR2 groups showed incomplete healing with persistent inflammatory cells and granulation tissue.The model group exhibited slow healing and marked scar tissue hyperplasia.The serum levels of IL-1β,IL-18and TNF-α in the combination group were significantly lower than in the model group(P<0.01). Conclusion Overexpression of CXCR2 and ADSCs significantly accelerated skin wound healing and reduced inflammation.

10 Preparation and Characterization of Carboxylated Graphene Oxide as a Pharmaceutical Carrier

Objective To explore the potential of graphene oxide as a drug carrier material,prepare functionalized graphene oxide and characterize it,providing an excellent carrier for achieving high drug loading,targeted delivery and controlled release of drugs. Methods Graphene oxide preparation by five-day oxidation method and covalently modified to become carboxylated graphene oxide.Infrared spectroscopy,transmission electron microscopy and other methods were used for characterization. Results The yield of carboxylated graphene oxide was 80.72%,and average particle size is 1510 nm.The sample had a distinct -OH absorption peak at 3393.37cm-1,and carboxyl -C=O peaks at 1732.01cm-1 and 1616.35cm-1,which were the stretching vibration peaks of the carbonyl C=O on the carboxylic acid at the edge of the carboxylated graphene oxide sheet.There was a carboxyl-C-O peak at 1072.42cm-1,which was the stretching vibration peak of C-O on the carboxylic acid at the edge of the carboxylated graphene oxide. Conclusion Compared with graphene oxide,carboxylated graphene oxide contains a larger amount of oxygen-containing functional groups in its structure,which is easy to combine with water molecules to form hydrogen bonds,thus providing an excellent drug carrier for achieving high drug loading,targeted drug delivery and controlled release.

14 Pharmaceutical Analysis of Atorvastatin in the Treatment of Elderly Patients with Coronary Heart Disease and Its Effects on Triglyceride and Total Cholesterol Levels in Patients

Objective To investigate the clinical efficacy of atorvastatin treatment in elderly patients with coronary heart disease(hereinafter referred to as "coronary heart disease")and its impact on triglyceride(TG)and total cholesterol(TC)levels. Method 68 elderly patients with coronary heart disease were selected as the observation subjects.According to the principle of random number table,the observation subjects were randomly divided into two groups,named control group and observation group respectively.The number of patients in each group was 34,and the treatment methods used were conventional treatment and conventional treatment plus atorvastatin treatment.The clinical total effective rate,blood lipid levels before and after treatment,and cardiac function indicators of the two groups were compared. Results Compared with the control group,the observation group showed significantly higher overall clinical effective rates(P<0.05);After treatment,the observation group showed lower levels of TG,TC,and low-density lipoprotein cholesterol(LDL-C)compared to the control group,and higher levels of high-density lipoprotein cholesterol(HDL-C)compared to the control group(P<0.05);Compared with the control group,the observation group had relatively lower left ventricular end systolic diameter(LVESD)and left ventricular end diastolic diameter(LVEDD),and relatively higher left ventricular ejection fraction(LVEF)compared with the control group(P<0.05). Conclusion Atorvastatin treatment has a significant effect on elderly patients with coronary heart disease,not only effectively regulating their blood lipid levels,but also improving their cardiac function indicators.

17 Effects of Pabolizumab on T Lymphocyte and Serum sMICA Levels in Patients with Non-small Cell Lung Cancer

Objective To observe the effects of pabolizumab on T lymphocyte subsets and serum soluble MHC-1 chain related protein A(sMICA)levels in patients with non-small cell lung cancer(NSCLC). Methods 140 patients with NSCLC were treated in our hospital.They were divided by random number table method,70 patients in the control group were treated with AP regimen,and 70 patients in the observation group were treated with palizol.The clinical efficacy of the two groups was compared,the peripheral blood T lymphocyte subsets(CD3+,CD4+,CD8+,CD4+/CD8+)were detected,the serological indicators[squamous cell carcinoma associated antigen(SCC-Ag),carcinoembryonic antigen(CEA),sMICA,Transforming growth factor β1(TGF-β1),Vascular endothelial growth factor(VEGF)] were detected,and the adverse reactions of the two groups were recorded. Results The DCR of the observation group was 84.29%,which was higher than the control group's 67.14%(P<0.05).After treatment,the levels of CD3+,CD4+,CD4+/CD8+ in observation group were and,which were higher than those in control group and.CD8+ level in observation group was,lower than that in control group,P<0.05.After treatment,the serum levels of SCC-Ag,CEA and sMICA in the observation group were and pg/mL.It was lower than that of control group,P<0.05.After treatment,the levels of TGF-β1 and VEGF in serum were and,which were lower than those in control group(29.03±4.12)μg/L and(377.14±35.98)ng/L,P<0.05.The incidence of adverse reactions was similar between the two groups(P>0.05). Conclusion Pabolizumab can regulate the level of T lymphocyte subsets,reduce the level of tumor markers and serum sMICA in non-small cell lung cancer NSCLC,and has good efficacy and safety.

20 The Value of Coronary CTA in the Evaluation of Acute Coronary Syndrome and the Degree of Coronary Stenosis was Analyzed

Objective To explore the value of coronary CT angiography(CTA)in evaluating acute coronary syndrome(ACS)and the degree of coronary stenosis. Methods A total of 87 suspected ACS patients were selected.All patients underwent coronary CTA and coronary angiography(CAG)to assess the degree of coronary stenosis.The results of CAG were used as the gold standard to analyze the diagnostic value of coronary CTA for ACS and the degree of coronary stenosis. Results Among the 87 suspected ACS patients,71 cases were detected by CAG,accounting for 81.61%;among them,14 had mild stenosis,32 had moderate stenosis,19 had severe stenosis,and 6 had complete occlusion.Based on the &quot;gold standard&quot;of CAG,the sensitivity,specificity,and accuracy of coronary CTA for diagnosing ACS were 98.59%,87.50%,and 96.55%,respectively,with good consistency compared to CAG results(Kappa=0.882).The diagnostic agreement rates for mild,moderate,severe,and complete occlusion in coronary CTA were and,respectively,with a total accuracy of 95.78%.Additionally,the consistency between coronary CTA and CAG results regarding the degree of coronary stenosis was good(Kappa=0.938). Conclusion Coronary CTA has high diagnostic value for ACS and can effectively assess the degree of coronary stenosis,providing Objective evidence for clinical diagnosis and treatment.

23 Therapeutic Effect of Recombinant Human Interferon a2b Spray on Hand Foot Mouth Disease in Children

Objective To observe the clinical effect of recombinant human interferon a2b spray in the treatment of hand foot mouth disease in children. Methods A total of 150 children with HFMD participated in this study.50 patients in Group A were treated with spray of recombinant human interferon a2b.In Group B,50 children were treated with ribavirin spray,while 50 children in Group C received placebo intervention.Analyze the differences in symptoms,duration of signs,treatment efficacy,and inflammatory factor indicators among the three groups. Results Children in Group A had a shorter time for symptoms and signs to disappear after treatment,a higher overall treatment effectiveness rate,and a decrease in inflammatory factor indicators.Comparing the values of the three groups,P<0.05. Conclusion Recombinant human interferon a2b spray can be the first choice for children with hand,foot and mouth disease,which can effectively shorten the duration of symptoms,improve immune function and reduce inflammatory indicators.

26 Observation on the Application Effect of Polyetheretherketone Removable Partial Denture in Patients with Prosthodontics

Objective To explore the application effect of polyetheretherketone(PEEK)material for removable partial denture(RPD)in patients with oral restoration. Methods 88 patients with oral cavity repair were randomly divided into two groups,44 cases in each group.The control group was treated with cobalt chromium alloy to make RPD,and the study group was treated with peek to make RPD.Periodontal indexes[plaque index(PLI),mobility(TM),periodontal probing depth(PD),sulcus bleeding index(BI)],satisfaction score,denture strength and bracket weight were compared between the two groups. ResultsBefore repair,there was no significant difference in PLI,TM,PD,Bi levels between the two groups(P>0.05);Compared with the control group,the PLI level in the study group was lower after 6 months of repair,with statistical difference(P<0.05),and there was no statistical difference in the levels of TM,PD and Bi between the two groups(P>0.05).Compared with the control group,the denture retention and stability,masticatory function,language,aesthetics,denture comfort,total score and other satisfaction scores of the study group were higher,with statistical difference(P<0.05).Compared with the control group,the denture strength score of the study group was lower,and the weight of the bracket was lighter,with statistical difference(P<0.05). Conclusion PEEK material in the treatment of RPD patients has achieved good repair effect,periodontal indicators have been effectively improved,patients' satisfaction with repair is high,and the strength of the repaired denture is strong,the weight of the bracket is light,which has a good prospect in clinical application.

29 Application of Abdominal and Vaginal Ultrasound and Contrastenhanced Ultrasound in Diagnosis and Differentiation of Uterine Fibroids and Adenomyosis

Objective To study the application value of abdominal and vaginal ultrasound and contrast-enhanced ultrasound in the diagnosis of hysteromyoma and adenomyosis. Methods To observe 121 cases of suspected hysteromyoma.All patients underwent abdominal and vaginal ultrasound and contrast-enhanced ultrasound examinations,with pathological examination results as the gold standard.Compare the efficacy of three diagnostic methods for uterine fibroids and adenomyosis. Results 121 patients with suspected hysteromyoma were pathologically diagnosed as hysteromyoma in 101 cases and adenomyosis in 20 cases.Sensitivity and specificity of contrast-enhanced ultrasound in diagnosing hysteromyoma were and higher than those of abdominal ultrasound and vaginal ultrasound 98.02% and 95.00%,respectively.The sensitivity and specificity of diagnosing adenomyosis were and,respectively,which were higher than those of abdominal ultrasound and vaginal ultrasound.The onset time,peak time and peak intensity of hysteromyoma patients are higher than those of adenomyosis patients(P<0.05). Conclusion The diagnostic efficiency of contrast-enhanced ultrasound is better than that of vaginal ultrasound and abdominal ultrasound,which can significantly improve the accuracy of differential diagnosis between hysteromyoma and adenomyosis.

32 Comparison of Anesthetic Effects and Postoperative Pain of Sacral Block and Subarachnoid Block under Ultrasound Guidance in Colorectal Surgery

Objective To investigate the application of ultrasound-guided sacral block and subarachnoid block in anorectal surgery. Methods A total of 60 patients undergoing anorectal surgery were selected and randomly divided into a control group(30 cases)and an experimental group(30 cases)using a sealed envelope method.The control group received subarachnoid block anesthesia,while the experimental group received ultrasound-guided sacral block anesthesia.The anesthetic effects,postoperative pain,total analgesic drug usage,analgesic pump pressurization frequency,and complications were compared between the two groups. Results The onset time for sensory and motor block in the experimental group was shorter than that in the conventional group,and the duration of anesthesia was longer(P<0.05).Compared to 6 hours post-surgery,both groups showed a decrease in VAS scores at 12 and 24 hours post-surgery,with the experimental group having lower VAS scores at 6,12,and 24 hours post-surgery(P<0.05).The total analgesic drug usage and analgesic pump pressurization frequency in the experimental group were also lower than those in the conventional group(P<0.05).The overall incidence of complications in the experimental group was lower than that in the conventional group(P<0.05). Conclusion Ultrasound-guided sacral block is effective in anorectal surgery,providing good anesthesia,reducing postoperative pain,and decreasing the use of analgesic drugs and the occurrence of complications.

35 Effect of Hysteroscopic Electroresection on Ovarian Function and Total Antioxidant Capacity Levels in Patients with Endometrial Polyps

Objective This study examines the impact of transcervical resection of polyps(TCRP)on ovarian function and total antioxidant capacity indicators in patients diagnosed with endometrial polyps. Methods This study enrolled 150 endometrial polyp patients randomly allocating them equally into control and TCRP groups(n=75 each)through blinded selection.The control group underwent hysteroscopic curettage while the TCRP group received transcervical resection,with comparative analysis of surgical parameters,PBAC scores,menstrual characteristics(cycle duration,recovery patterns),endometrial thickness,hemoglobin levels,hormonal profiles(FSH,LH,PR,ER expression),along with complication and recurrence rates. Results The TCRP group demonstrated superior surgical outcomes with significantly shorter operation time and reduced intraoperative blood loss(P>0.05),though requ-iring marginally longer postoperative hospitalization days(P<0.05).Both groups showed marked clinical improvement postoperatively(P<0.05),with the TCRP group exhibiting significantly better PBAC scores,shorter menstruation duration, thinner endometrium,and lower Hb levels versus controls(P<0.05).While the TCRP group had prolonged menstrual recovery time,heavier initial flow and extended bleeding duration(P<0.05),it showed significantly lower postoperative hormone receptor expression and gonadotropin level(P<0.05).Notably,the TCRP group achieved substantially reduced complication and recurrence rates(P<0.05). Conclusion TCRP demonstrates significant therapeutic efficacy for endometrial polyps,effectively reducing menstrual blood loss,endometrial thickness,and hormone levels while promoting menstrual recovery,despite requiring slightly longer hospitalization,with the added advantage of minimizing polyp recurrence rates.

39 Effect of Metal Precrown and Resin Filling on Dental Caries in Children's Deciduous Teeth

Objective To investigate the effect of resin filling and metal precrown on dental caries in children's deciduous teeth. Methods According to random number table method,78 children who underwent molar dental caries were divided into reference group and study group.Selected 39 patients in each group were included.The reference group underwent traditional resin-filled restoration,and the study group underwent metal precrown restoration.The repair effect,occlusal ability,masticatory efficiency and complications were compared between the two groups. Results The excellent repair rate of the study group was,which was higher than that of the reference group,with statistical difference(P<0.05).There was no significant difference in occlusal ability and masticatory efficiency between the two groups before repair(P>0.05).The maximum occlusal area,masticatory efficiency and masticatory force of the two groups were higher than those before repair 6 months after repair,while the maximum occlusal area of the study group was mm2,the maximum occlusal force was Ibs and the masticatory efficiency was.Higher than in the reference group,there were statistical differences(P<0.05).The complication rate of the study group was lower than that of the reference group(P<0.05).The incidence of post-repair complications in the study group,which was lower than that in the reference group,with statistical difference(P<0.05). Conclusion The high success rate of metal precrown restoration for deciduous molar proximal caries can effectively improve the occlusal ability of children,help children chew food more efficiently,effectively improve chewing efficiency,and significantly reduce the incidence of complications such as restoration loosening,loss and secondary caries,which has high application value in the treatment of children oral diseases.

43 Early Treatment Response and Prognostic Influences of Childhood ALL

Objective To analyze the early treatment response and prognostic factors of children with acute lymphoblastic leukemia(ALL). Methods 80 cases of children with ALL in our hospital were selected as experimental subjects,and all the subjects were divided into 17 cases in the poor prognosis group(relapse or death)and 63 cases in the good prognosis group(no relapse or death)according to their prognosis.Sex,age,immunophenotyping(B-cell type,T-cell type),bone marrow morphology(M1,M2,M3),initial platelet count(PLT),fusion gene(negative,benign risk,poor risk),response to prednisone test(good response,poor response),and microscopic residual lesions(MRDs)on the 19th and 46th day of induction chemotherapy were collected in the 2 groups,and were compared.early treatment response of the 2 groups and analyze the factors influencing the prognosis of ALL. Results The incidence of poor prednisone test response and MRD positivity on the 46th d of induction chemotherapy were higher in the poor prognosis group than in the good prognosis group and(P<0.05);initial WBC,fusion gene,prednisone test response,and MRD on the 46th d of induction chemotherapy were the single factor affecting the prognosis of ALL(P<0.05);logistic regression analysis showed that initial WBC≥100×109/L,poor risk of fusion gene,poor response to prednisone test,and high MRD positivity on the 46th d of induction chemotherapy were risk factors affecting the poor prognosis of children with ALL(P<0.05). Conclusion Early treatment response can assess the prognostic outcome of children with The prognostic effect of ALL,and the high initial WBC and poor response to prednisone test are risk factors affecting the poor prognosis of children with ALL,and the clinic can take active preventive measures according to the relevant factors to improve the survival rate of children.

46 Clinical Value of Dipyridamole Combined with Standard Regimen in the Treatment of Kawasaki Disease in Children

Objective To explore the clinical value of dipyridamole combined with standard regimen in the treatment of Kawasaki disease in children. Methods 131 children with Kawasaki disease were prospectively selected.According to the condition of children,different treatment schemes were selected and grouped as follows:Group A(65 cases,treated with immunoglobulin and aspirin)and Group B(66 cases,treated with dipyridamole in addition to immunoglobulin and aspirin).The treatment period was 6-8 weeks.The indexes between groups and within groups were contrasted and analyzed. Results The total effective rate of Group B was higher(compared to Group A),P<0.05.The relief time of various clinical symptoms in Group B was shorter(compared to Group A),P<0.05.The levels of CD8+in peripheral blood increased in both groups(compared to before treatment and after treatment),and Group B was higher(compared to Group A,after treatment);Two groups of peripheral blood CD3+ CD4+、CD4+/CD8+、PLT.The plasma Fbg levels decreased(compared to before treatment and after treatment),and group B had lower levels(compared to group A,after treatment),both P<0.05. Conclusion On the basis of the standard scheme,dipyridamole combined with dipyridamole is effective in treating Kawasaki disease in children.which can effectively alleviate clinical symptoms,improve immune function and coagulation function.

49 The Effect of Single Port Laparoscopic Appendectomy in the Treatment of Acute Complex Appendicitis in Children

Objective To investigate the efficacy of single port laparoscopic appendectomy(SILA)in the treatment of acute complex appendicitis(ACA)in children. Method 100 children with ACA who received treatment were selected and divided into two groups based on different surgical methods:the traditional laparoscopic appendectomy group(LA group,49 cases)and the umbilical single port laparoscopic appendectomy group(SILA group,51 cases). Results And analyze and compare the surgical related indicators,pain levels,inflammatory factor levels,and incidence of complications between the two groups.There was no significant difference in surgical time,intraoperative blood loss,and defecation time between the two groups(P>0.05),and the hospital stay in the SILA group was shorter than that in the LA group(P<0.05).On postoperative day 1 and 2,the pain scores in the SILA group were lower than those in the LA group(P<0.05).On postoperative day 1 and 2,the levels of serum CRP,PCT,and WBC in both groups showed a trend of first increasing and then decreasing.During this period,there was no significant difference in the levels of CRP,PCT,and WBC between the two groups on postoperative day 1(P>0.05),while on postoperative day 3,the SILA group was lower than the LA group(P<0.05).There was no significant difference in the incidence of complications between the two groups(P>0.05). Conclusion Compared with LA,SILA can significantly shorten hospitalization time,reduce postoperative inflammatory response,and effectively alleviate postoperative pain in the treatment of ACA children,with higher safety and faster postoperative recovery.
