Anti Fibrosis Effect of α-MSH on Bleomycin Induced Pulmonary Fibrosis in Mice

Abstract:Objective To observe the effect of α-MSH on the fibrosis factor type I collagen,key nuclear factor in TNF-α and immune-regulation pathway,we initially investigated whether α-MSH has an anti-fibrotic effect in different periods of pulmonary fibrosis. Methods The mouse lung fibrosis model was constructed by bleomycin tracheal drip method,and α-MSH was treated in the first,second and third weeks of animal model to detect the expression of type I collagen,TNF-α mRNA and production of NF-κB protein,and the generation of TNF-α in peripheral blood and alveolar lavage fluid. Results α-MSH could significantly inhibit the expression of type I collagen and TNF-α gene.The production of p-NF-κB protein in the mouse model of bleomycin-induced pulmonary fibrosis could inhibit by α-MSH,which had obvious anti-inflammatory and anti-fibrosis effect.Furthermore α-MSH began to exert this effect in the early stage of the fibrosis,and had the characteristics of long maintenance time.It suggests that α-MSH has an important role in the early immune action mechanism of pulmonary fibrosis. Conclusion α-MSH may be a new target for early treatment of IPF and has great value for early prevention and treatment of IPF.
