Abstract:Objective To analyze the clinical value of real-time fluorescent quantitative PCR(FQ-PCR) for detection of Mycobacterium tuberculosis(MTB) DNA in paraffin tissue in screening tuberculosis(TB). Method 118 TB patients admitted to our hospital from January 2017 to December 2019 were selected, and tissue specimens were collected, embedded in paraffin, and subjected to pathological examination;Furthermore,FQ-PCR and acid fast staining of tissue sections were used to detect MTB-DNA,and the positive detection rates of the three methods were compared. Result Among the paraffin embedded tissues of 118 selected patients,FQ-PCR,acid fast staining, and histopathological methods were used to detect MTB positivity in 70,50,and 65 cases, respectively, with positive rates of 59.32%,42.37%,and 55.08%,respectively.The positive rates of the three methods were significantly different(P<0.05).Compared with the acid fast staining method, the positive detection rate of MTB in lung, pleural, lymph node biopsy, and other tissue specimens is higher,FQ-PCR、 The histopathological method showed higher detection rates(P<0.05),while the positive detection rate of FQ-PCR in lung biopsy specimens was higher than that of histopathological method(P<0.05). Conclusion The use of FQ-PCR for detecting MTB-DNA in paraffin tissue is more effective than acid fast staining and tissue pathology methods in lung biopsy. It can be used as an important tool for screening TB and provide support for tissue pathology diagnosis.
