Lu Jianjun, Yu Qin, Chen Lixiong
DOI: 10.3969/j.issn.1001-8131.2024.01.001
Objective This research was intended to exploring the effect and mechanism of kinase(NLK)and paxillin on proliferation, migration, invasion, and epithelial mesenchymal transition(EMT)of bladder cancer cells, and providing theoretical basis for basic research, clinical prevention and treatment of bladder cancer. Methods The expression of NLK in bladder cancer tissue was analyzed by bioinformatics analysis. The proliferation, migration and invasion of bladder cancer cells were detected by EdU staining assay, clone formation assay, and transwell assay.RNA pull down assay was used to detect the interaction between NLK and paxillin in bladder cancer cells.Western blot assay was used to detect the expressions of NLK,p-paxillin,and EMT-related proteins in bladder cancer cells. Results NLK and p-paxillin were highly expressed in bladder cancer;NLK interacted with paxillin;NLK downregulation inhibited the proliferation, migration, invasion,EMT,and p-paxillin expression of bladder cancer cells;paxillin overexpression reversed the effect of silencing NLK on proliferation, migration, invasion,EMT,and p-paxillin expression in bladder cancer cells. Conclusion NLK promoted the proliferation, migration, invasion and EMT of bladder cancer cells by regulating paxillin, providing theoretical basis for basic research, clinical prevention and treatment of bladder cancer.

DOI: 10.3969/j.issn.1001-8131.2024.01.002
Objective To explore the relationship between cerebral blood flow, serum neurotrophic factor-3(NT-3),brain-derived neurotrophic factor(BDNF)and functional recovery in patients with acute cerebral infarction(ACI)and hemiplegia after intervention. Methods A total of 60 patients with ACI admitted to the hospital. After admission, all underwent routine treatments(anti-coagulation,anti-platelet,lipid regulation, removal of oxygen free radicals, promoting circulation and removing stasis)and comprehensive rehabilitation therapy. After 4 weeks of continuous intervention, cerebral blood flow [anterior cerebral artery(ACA),middle cerebral artery(MCA)],levels of serum NT-3 and BDNF,and functional recovery [National Institutes of Health Stroke Scale(NIHSS),Fugl-Meyer Assessment Scale(FMA)] were compared before and after treatment. Results After treatment, cerebral blood flow of ACA and MCA was increased(P<0.05).After treatment, level of serum NT-3 was decreased, while BDNF level was increased(P<0.05).After treatment,NIHSS score was decreased, while FMA score was increased(P<0.05).ACA,MCA and BDNF were negatively correlated with NIHSS score, while positively correlated with FMA score(P<0.05).NT-3 was positively correlated with NIHSS score, while negatively correlated with FMA score(P<0.05). Conclusion The cerebral blood flow, levels of serum NT-3 and BDNF significantly improve in patients with acute cerebral infarction and hemiplegia after intervention, which can better reflect the recovery level of limb function and motor function.

DOI: 10.3969/j.issn.1001-8131.2024.01.004
Objective To explore the predictive value of CK20,CDX2,CK7 tissue expression levels for postoperative curative effect and prognosis of colorectal cancer. Methods The basic data of 60 patients with colorectal cancer were collected and analyzed retrospectively. All patients underwent radical resection of rectal cancer.Immunohistochemical SP method was used to detect the expression of CK20,CDX2 and CK7 in tumor tissues, and the patients were followed up for one year.By reexamining colonoscopy, the patients were divided into a good prognosis group(n=26)and a poor prognosis group(n=34)according to whether the tumor recurred or metastasized after operation.Univariate analysis and multivariate Logistic regression analysis were used to analyze the influencing factors of the prognosis of colorectal cancer patients. Results Compared with before operation, after operation, the positive expression rates of CK20 and CK7 in colorectal cancer(80.00%,83.33%)decreased,but the positive expression rate of CDX2(76.66%)increased(P<0.05).The Logistic regression model showed that the mass size(P=0.034,OR=1.411),CK20(P=0.024,OR=1.850),CDX2(P=0.013,OR=1.540),CK7(P=0.012,OR=1.540). Conclusion The expression levels of CK20,CDX2 and CK7 play an important role in the occurrence and development of colorectal cancer, and have certain clinical application value in evaluating the postoperative curative effect and prognosis of colorectal cancer.

Objective To investigate the risk factors of delayed bleeding after endoscopic submucosal dissection(ESD)for digestive tract lesions. Methods Clinical data of 160 patients with digestive tract lesions treated with ESD were retrospectively analyzed, and relevant clinical, endoscopic and pathological data of the patients were analyzed.Patients were divided into bleeding group and non-bleeding group according to whether delayed bleeding occurred. The above data were compared between the two groups, and the risk factors of delayed bleeding after ESD were summarized. Results There was no significant difference between the two groups in terms of age, gender, complications, and whether they had taken anticoagulant drugs in the past(P>0.05).The size of lesions and the proportion of surface ulcers in bleeding group were higher than those in non-bleeding group(P<0.05).Total resection of the two groups was compared(P>0.05).The operation time and the proportion of obvious bleeding in the bleeding group were higher than those in the non-bleeding group(P<0.05).Lesion size, surface ulcers, operation time and obvious bleeding during operation were all related factors affecting delayed bleeding after digestive tract lesions(P<0.05). Conclusions Patients with digestive tract lesions with large lesions, surface ulcers, long operation time and obvious bleeding during operation have higher risk of postoperative delayed bleeding, and should be treated as soon as possible.

Objective To explore the effects of different atomization dosing sequences on the asthmatic remission time of Infantile Asthmatic Pneumonia, and to provide theoretical basis for improving nursing operation process. Method A total of 475children with asthmatic pneumonia who met the inclusion criteria were randomLy divided into two groups. The children in both groups were treated according to the consensus of experts in the diagnosis and treatment of children's asthmatic pneumonia. The observation group was treated with atomization inhalation, first adding 2mL normal saline to lubricate the wall of the atomization cup and shaking it well, and then adding other atomization drugs. In the control group, conventional atomization drug dosing method was used.To compare the time of beginning relief of wheezingand the time of complete disappearance of wheezing, the average atomization time and tube plugging time of the first atomization cup during use. Results The time of remission and complete disappearance of wheezing in the observation group was earlier than those in the control group(P<0.05).The average atomization time of the observation group was shorter than that of the control group, and the use time of the atomization cup was significantly prolonged, with statistical significance(P<0.01). Conclusion During atomization inhalation treatment, add normal saline to lubricate the inner wall of the atomization cup, which can shorten the atomization time of children, ensure the atomization effect, significantly shorten the wheezing time of children, and prolong the service life of the atomization cup.

134 Research Progress on the Screening Methods For Hydroxychloroquine Retinal Toxicity

DOI: 10.3969/j.issn.1001-8131.2024.01.046
Long-term excessive use of hydroxychloroquine (HCQ) can lead to retinal damage, a well-established ocular side effect that can cause severe and irreversible central vision loss in advanced stages. However, early lesions are usually asymptomatic and therefore difficult to detect. With the development of automated visual field and retinal technology, early lesions can be detected before significant visual impairment occurs. Therefore, sensitive and effective screening for early retinopathy is essential. This article will focus on reviewing the screening methods for retinal toxicity in HCQ in order to improve the understanding of retinal toxicity screening in HCQ.
